摘要 目的构建孕鼠慢性牙周炎模型,探讨孕鼠慢性牙周炎胎盘组织、血清及羊水中基质金属蛋白酶-3的表达。方法20只Wistar雌鼠随机分为两组:正常组(n=8)、实验组(n=12),各组接受相应的建模处理。观察其牙周组织的病理改变,实时荧光定量PCR分析胎盘组织中基质金属蛋白酶-3的表达,酶联免疫吸附实验检测血清及羊水基质金属蛋白酶-3含量。结果实验组牙周组织改变:牙槽骨吸收,牙周袋形成,组织病理见有大量的破骨细胞、附着丧失。实时荧光定量PCR及酶联免疫吸附试验检测基质金属蛋白酶-3,两组有差距有统计学意义。结论慢性牙周炎可以促进基质金属蛋白酶-3的合成及分泌,并进入血循环通过胎盘屏障,增加不良妊娠的机率。 Objective To detect and analyze the espression of matrix metalloproteinase-3 in blood - serum and amniotic fluid of construction of pregnant rats chronic peridontitis model. Methods 20 wistar female rats were randomly divided into two groups: the normal group (n = 8 ) and the experimental group (n = 12). Every group accepted the corresponding measurements to establish an animal model. To observe the periodontal tissues and pathology changes, the expression of matrix metalloproteinase-3 in placenta tissue was tested by Real-time PCR. ELISA was employed to detect the content of matrix metalloproteinase-3 in blood-serum and amniotic fluid. Results In frontal-resorption and periodontal pocket, a large number of osteoelasts and attachment loss were found in histopatbology. The content of matrix metallo-proteinase-3 was obviously different in two sets ( P 〈 0.05 ). Concluctions The chronic periodontitis may high-regulate the expression of matrix metalloproteinase-3 into blood circulation through placental barrier.
