摘要 目的:研究干扰RNA质粒载体对肿瘤细胞的作用。方法:制备了针对FGF3基因的干扰RNA质粒载体(pRNATU6.1/NeoGFP-FGF3),并选用大肠癌细胞株HT-29为实验对象,流式细胞仪观察干扰RNA质粒载体对HT-29细胞周期和凋亡活性的影响。结果:细胞周期分析发现转染pRNATU6.1/NeoGFP-FGF3后,HT-29细胞增殖能力减弱;细胞凋亡实验表明干扰RNA质粒载体有明显的促进肿瘤细胞凋亡作用。结论:干扰RNA质粒载体能明显抑制HT-29肿瘤细胞的增殖,并且能显著促进细胞凋亡。 Objective:To study the effect of pRNATU6.1/NeoGFP - FGF3 on the HT - 29 cells. Methods:pRNATU6.1/NeoGFP - FGF3 was designed to FGF3 gene and colorectal cancer cell line HT - 29 was used for the experiment,the effect of pRNATU6.1/NeoGFP -FGF3 on the cell cycle and apoptosis of HT -29 cell was observed by flow cytometry. Results: more cells treated with pRNATU6.1/NeoGFP - FGF3 stayed in G2/M stage and the cell pro- liferation rate decline, compared to control ceils, pRNATU6.1/NeoGFP - FGF3 increased the apoptosis activity of HT29 cells. Conclusion : SiRNA vector (pRNATU6.1/NeoGFP -FGF3 ) can affect the cell cycle, promote the apoptosis and inhibit the proliferation of HT29 cell significantly.
