摘要
目的通过单个胚胎荧光定量RT-PCR的方法分析不同质量和来源的胚胎与IGF-Ⅱ表达量之间的关系。方法收集不适合移植及冷冻保存的胚胎,收集胚胎评分资料和所属患者资料。运用荧光定量RT-PCR方法测定其IGF-Ⅱ表达量。结果人类植入前胚胎形态学评分越高,其IGF-ⅡmRNA的表达量越高;年龄较轻和妊娠的病人中其胚胎中IGF-Ⅱ的基因表达量较高。结论荧光定量RT-PCR技术可用于检测单个植入前胚胎的mRNA含量;IGF-II可作为胚胎质量的标记物;IGF-II基因表达量和胚胎的形态学评分呈正相关。
Objective: To examine the suitability of fluorescence monitored reverse transcription (RT -PCR) for the study of single preimplantation embryo's gene expression and to approach the relationship between IGFs content and the quality of human embryos. Methods : Human preimplantation embryos unsuitable to transfer or freeze were collected. Real time PCR was performed to detect the IGF - Ⅱ of a single human preimplantation embryo. Results : The amount of IGF -Ⅱ detected in good quality embryos, however, was significantly greater than in poor embryos. The younger the patient was, the higher the amount of IGF - Ⅱ in her embryos. The pregnant patient has higher amount of IGF - Ⅱ . Conclusion : Fluorescence monitored RT - PCR from individual embryos can be used as a sensitive tool for the study of early developmental processes of human embryo. The amount f IGF - Ⅱ detected in good quality embryos, however, was significantly greater than in poor embryos. Expression of IGF - Ⅱ may be a potential marker for embryo quality . The intensity of gene expression was positively correlated with the morphology of embryos especially the cell numbers.
