摘要 目的探讨地塞米松对体外培养成骨细胞生长的影响。方法采用新西兰大白鼠股骨骨髓单个核细胞行体外培养成骨细胞,根据加入成骨细胞培养液中相同浓度(1×10^-1mol/L)地塞米松的不同剂量将其分为20μl、30μl、40μl组,并设对照组(培养液中无地塞米松),分别采用倒置显微镜和瑞氏一姬姆萨染色、碱性磷酸酶染色、苏丹Ⅳ染色观察活细胞形态、数量及细胞内成分。结果地塞米松各组抑制细胞增生的作用显著优于对照组(P〈0.01);对照组骨髓单个核细胞均可转变为成骨细胞,而地塞米松各组均可抑制骨髓单个核细胞向成骨细胞分化,并促使其脂肪变性,尤以30μl和40μl组最强。结论地塞米松可抑制体外培养的骨髓单个核细胞向成骨细胞转化,且作用与其剂量呈正相关。 Objective To study the effect of dexamthasone on the grouth of the osteoblast in vitro. Methods On the basis of culturing osteoblast with mononucleate cells that were in bone-marrow in femoral bone in mouse. Mononucleate cells in cultures were treated with the same concentrations and differ dosage dexamethasone (10-7mol,20,30,40μl).We studied the effect of dexamethasone on mononucleate cells with contrast group that didn't contain dexamthasone in the medium.The cells were distinguished with invert microscopy,Wright-Geimsa mixture stain,alkaline phosphatase (ALP) stain (Gomor's) and SudanIVstain. Results The test showed that dexamethasone groups can keep from the grouth of the cells (P〈0.01),and prevent the cells from differentiating into the osteoblasts and bring into fatty degeneration, specially 30-40μl dexamethasone groups. Conclusion Dexamethasone prevent mononucleate cells from differentiating into the osteoblasts and was positive correlation with dosage dexamethasone in vitro.
