人IL-4选择性剪接变异体(IL-4δ2)cDNA的克隆和序列测定

在对多种细胞因子的激活条件进行对比实验的基础上，我们成功地构建了人外周血激活淋巴细胞ｃＤＮＡ单链库。并采用ＩＬ－４特异引物和ＲＴ－ＰＣＲ技术，在从此库中克隆出了ＩＬ－４ｃＤＮＡ的同时，也获得了其选择性剪接变异体（ＩＬ－４δ２）ｃＤＮＡ。测序结果表明，所克隆的ＩＬ－４δ２ｃＤＮＡ含有成熟ＩＬ－４蛋白的外显子１，３，４编码区和前导肽编码序列，唯编码１６个氨基酸的外显子２在转录后被剪接掉。这一研究为今后进一步探究细胞因子选择性剪接变异体的工作奠定了基础。

With RT PCR and IL 4 specific primers, we obtained the hIL 4δ2 cDNA that was cloned from the first strand cDNA library of activated lymphocytes, which were rich in cytokine cDNA. The amplified products of hIL 4δ2 cDNA were cloned into pGEM 3zf(+) vector and were sequenced with dideoxynucleotide termination method. The results showed that the gene comprises exons 1, 3 and 4 of IL 4 in an open reading frame, but the second exon which encoding 16 amino acids was deleted by alternative splicing.
