放射线和加热诱导CNE-2细胞系发生凋亡:几种检测方法的比较 被引量:4

摘要 比较了荧光显微镜观察法,流式细胞仪检测法及TdT末端标记法检测高能X线及加热诱导人鼻咽癌细胞系的凋亡指数。结果发现CNE-2细胞系受照2、4、8、12及20Gy后三种检测方法的凋亡指数荧光法分别为15%、20%、31%、38%及48%;流式法为16.5%、20.1%、33%、42.3%及47.5%;TdT法为24.6%、30.0%、40.6%、50.1%及58.7%。我们使用TdT法作为加热诱导凋亡的检测方法,发现于41℃培养0.5、1、1.5、2.5h后凋亡指数分别为16.7%、24.5%、30.7%及43%。结果提示TdT末端标记法是比较敏感的检测方法,CNE-2细胞系是对放。 Purpose: To compare various methods of identification of apoptosis induced by irradiation and to investigate the apoptotic response of a cell line of nasopharyngeal carcinoma (CNE 2). Methods: CNE 2 cell line was cultured as usual. Irradiation and heating were applied with a 6MV X ray accelerator and thermotat; fluorescence microscopy, flow cytometry and TdT terminal labelling technics were used to identify apoptotic features. Results: It was shown that the apoptotic rates of CNE 2 after irradiation of 2, 4, 8, 12 and 20 Gy were 15%, 20%, 31%, 38% and 48% by fluorescence method; 16.5%, 20.1%, 33%, 42.3% and 47.5% by flowcytometry method; and 24.6%, 30%, 40.6%, 50.1% and 58.7% by TdT method respectively.It was found that heating could also induce apoptosis and that at 0.5, 1, 1.5 and 2.5 hours after being heated the apoptotic indexes were 26.7%,34.5%,40.7% and 53% respectively. Conclusion:CNE 2 cell line is sensitive to irradiation and hyperthermia. Thus, it might be useful in individualized treatment planning.TdT end labelling method is sensitive in identifying apoptosis and distinguishing apoptosis from cell necrosis.
