红花注射液联合多西环素对兔急性脊髓损伤后MMP-2、12表达及神经功能的影响 被引量:6

摘要 目的:探讨兔急性脊髓损伤后应用红花注射液联和多西环素(Doxycycline)对基质金属蛋白酶-2、12(MMP-2、12)表达及神经功能的影响。方法:家兔随机分组,分别为脊髓损伤后应用生理盐水对照组(A组)和红花注射液联合多西环素的治疗组(B组),损伤后不同时间点取材,HE染色观察损伤脊髓组织病理变化;免疫组化染色检测MMP-2及Real-TimePCR法检测MMP-12的表达变化;采用Tarlov评分评价神经功能。结果:HE染色镜检发现脊髓组织病理学改变B组明显轻于A组。2组均发现MMP-2、12的表达,MMP-2、12表达A组>B组(P<0.05)。B组神经功能改善优于A组。结论:红花注射液联合多西环素能抑制家兔急性脊髓损伤(ASCI)后MMP-2、12表达,促进神经功能恢复。 Objective: To investigate the effect ofdoxycycline and HongHua injection on expression of MMP-2,12 and nerve function after acute spinal cord injury (ASCI) in rarebits. Methods: Rarebits were randomly divided into a control group of doxycycline (A group) which received normal saline and a treatment group of HongHua injection (B group) which received HongHua injection. After operation, the rarebits were sacrificed at 8 h, 24h, 48h, 72h and 7d .The tissue sections were stained with hematoxylin and eosin (HE) and the injured spinal cord tissue was evaluated by immunohistochemistry and real-time PCR method. Results: The histopathology changes in spinal cord tissue observed with HE staining microscopic examination in group A was worse than that in group B. The expression of MMP-2,12 were observed in two groups and the expression of MMP-2,12 in group A was higher than that in group B (P〈0.05). Conclusion: This study showed that doxycycline and HongHua injection can inhibit the expression of MMP-2,12 after ASCI in spinal cord injury rarebits and enhance the recovery of nerve function.
