摘要
目的通过建立裸鼠人胃癌MKN-45原位移植瘤模型,观察消痰散结方对胃癌组织核转录因子NF-κB P65蛋白表达的影响,探讨消痰散结方抑制胃癌细胞增殖的作用机制。方法采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植模型,随机分成模型组,消痰散结低、中、高剂量组,5-Fu组,每组8只,观察消痰散结方干预对胃癌组织NF-κB P65蛋白表达的影响。结果①消痰散结方低、中、高剂量组抑瘤率分别为36.62%、44.82%、58.11%,呈现出剂量依赖效应,且消痰散结方高剂量组抑瘤率高于5-Fu组(51.82%),差异无统计学意义;②消痰散结方低剂量组NF-κB P65蛋白的阳性表达与模型组相比无统计学差异(P>0.05),消痰散结方中、高剂量组、5-Fu组NF-κB P65蛋白的阳性表达与模型组相比P<0.05,有统计学意义,但组间相比无统计学意义。结论消痰散结方对裸鼠人胃癌原位移植瘤有较好的抑制肿瘤生长作用,揭示其发挥作用的关键与抑制NF-κB P65蛋白的表达有关,从而阻断胃癌发展,控制肿瘤复发与转移。
Objective To discover the mechanism of Xiaotan Sanjie Fang (XTF), a traditional Chinese medicinal empirical Prescription for curing gastric cancer in inhibiting proliferation of gastric cancer cells and observe the influence of Xiaotan Sanjie Fang intervening the expression of human gastric cancer MKN-45 Nuclear transcription factor NF-κBP^65 protein by establishing the nude mice human gastric cancer MKN-45in situ transplantation tumor model. Methods Using OB glue paste method, the nude mice human gastric cancer MKN-45in situ transplantation tumor model was established, based on which 40 animals were randomly divided into 5 groups (8 mice/every group) : the blank model group, low-dose XTF group, intermediate-dose XTF group, high-dose XTF group, and 5-Fu group. Observing the influence of Xiaotan Sanjie Fang intervening the expression of human gastric cancer MKN-45 Nuclear transcription factor NF-κBP^65 protein . Results (1)The inhibition tumor ratioes of the low, mid, high-dose of Xiaotan Sanjie Fang (XTF) group were 36.62% ,44.82% ,58.11%, respectively, which show dose-dependent effect, and the ratio of highdose Xiaotan Sanjie Fang group was higher than 5-Fu group (51.82%) ,differences have not statistical meaning ;(2)There were not statistical differences between the expression of NF-κB P^65 protein in low -dose Xiaotan Sanjie group and normal NS group ( P 〉 0.05 ). There was a statistical meaning by comparing the mid, high- dose Xiaotan Sanjie group ,5-Fu group with the normal NS group( P 〈0.05 ), but there were not statistical meanings among the different groups. Conclusion XTF can inhibit the tumor growth of human gastric cancer in a nude mice in situ tumor transplantation,Reveal its key role to inhibit of NF-κBP^65 protein expression, thus block the development of gastric cancer,Control of tumor recurrence and metastasis.
