摘要
目的观察CD36功能肽段YRVRYLAKENITQDPEDH(93-110)对小鼠实验性矽肺纤维化的阻抑作用。方法将小鼠按体重随机分为4组:生理盐水组、SiO2模型组、CD36功能肽段组(93-110)、CD36对照肽段组(208-225)。采用暴露式气管内注入法染尘,分别于第7、第14、第21天处死动物,HE、VG染色进行肺组织病理学形态观察,采用碱裂解法测定肺组织中羟脯氨酸含量。结果染尘7 d,病理检查表明,CD36功能肽段组小鼠肺组织肺泡炎症较SiO2模型组和CD36对照肽段组轻。小鼠羟脯氨酸含量在各组间的差异也没有统计学意义(P>0.05)。在染尘后14 d、21 d时,CD36功能肽段组小鼠肺组织纤维化程度明显轻于SiO2模型组和CD36对照肽段组小鼠,羟脯氨酸含量明显低于SiO2模型组和CD36对照肽段组,差异有统计学意义(P<0.05)。结论CD36功能肽段对小鼠实验性矽肺纤维化具有明显的抑制作用。
Objective To observe the inhibitory effect of CD36 synthetic peptide(93-1100)on fibrosis of silicotic mice. Methods Mice were randomly divided into four groups: the saline control group, SiO2 model group, CD36(93-110)group and CD36(208-225) group. 6 - 8 mice of each group were sacrificed after 7, 14 and 21 days respectively after instillation. Pathomorphology of silicotic lungs was observed using hematoxylin eosin(HE)and van Gieson stains(VG)dyeing. Changes of hydroxyproline contents in pulmonary tissue were measured. Results The inflammatory or fibrotic lesion in the lungs of CD36(93-110)group was lower than that of CD36 (208-225)group. The hydroxyproline contents of four groups at 7 days after the instillations had no significant differences. The fibrotic degree in silicotic mice lungs and hydroxyproline contents of pulmonary tissue were much lower in CD36 synthetic peptide treated group than those of SiOz model group and CD36 control peptide group at 14 d and 21 d(P〈0.05). Conclusions CD36 synthetic peptide(93-110)might have obvious inhibitory effect on lung fibrosis in silicosis mice.
