摘要
以人白血病HL-60细胞为实验对象,利用3H-脱氧胸苷(3H-TdR)掺入实验,JAM实验法及流式细胞仪观察槲皮素对HL-60细胞DNA和细胞周期的影响。结果显示,槲皮素15μmol/L~120μmol/L可显著抑制HL-60细胞的DNA合成及诱发DNA链断裂;槲皮素主要是阻滞G1期向S期移行,使G1期细胞蓄积,S期细胞减少。槲皮素的作用呈明显的剂量依赖关系。
Effects of quercetin, a flavonoid found in many plants, on DNA and cell cycle of human promyelocytic leukemia cells (HL60) were studied. Assay to 3H-TdR incorporation, JAM test and flow cytometric analysis were undertaken. Quercetin, 15~120 μ mol/L decreased the syntheses of DNA, degraded their DNA into small fragments, mainly blocked to progress through S-phase and arrested at G1 phase of HL60 cells in a concentration-dependent manner. Results suggested that antitumor action of quercetin may be related to its inhibition of DNA syntheses and initiation of DNA damage.
