摘要
建立了格列喹酮血药浓度的测定方法。该法使用SpherisorbC18色谱柱,以乙腈-水(6535)为流动相,流速为1.0ml/min,检测波长为310nm,灵敏度为0.001AUFS。线性范围为50~2000ng/ml,方法回收率为95%以上,日内、日间误差小于7.0%。
A HPLC method was established to determine the plasma concentration of gliquidone The method utilized a Spherisorb C 18 column, CH 3CN H 2O (65∶35) as mobile phase with the flow rate of 1 0 ml/min and 310 nm as detective wavelength at a sensitivity of 0 001 AUFS Gliquidone was abstracted by 4 ml methylene dichloride from 1 ml plasma, then 3 ml methylene dichloride was transfered to another tube and dried by air at 50℃, the residue was dissolved by 100 μl mobile phase and 20 μl was injected to the column The standard curve showed a linearity in the range of 50 ̄2000 ng/ml The recovery of the method was over 95% The Intra day and Inter day RSD were less than 7 0%
