^(99m)Tc-抗神经母细胞瘤单抗在荷瘤裸鼠体内的放免显像 被引量:2

Distribution and Immunoscintigraphy of ^(99m)Tc-Monoclonal Antibody Against Neuroblastoma in Nude Mice

摘要 小儿神经母细胞瘤严重危害儿童的健康,放射免疫显像方法为肿瘤的早期诊断提供依据。利用杂交瘤技术制备抗神经母细胞瘤单克隆抗体,用"预锡"直接标记法标上^(99m)Tc,在荷人神经母细胞移植瘤裸鼠体内获得肿瘤的放射免疫显像,并计算各脏器的 ID%和 T/NT 值。结果显示单克隆抗体的标记率达97%,免疫活性为85%,标记的单抗在裸鼠瘤体内随时间延长呈浓聚趋势,瘤体显像渐趋明显,ID%值上升,而其它组织的 ID%值则下降,T/NT 值升高。对照组则无此现象。说明研制的抗神经母细胞瘤单克隆抗体标上^(99m)Tc 后对荷人神经母细胞移植瘤有定位显像作用。 Neuroblastoma(NB) endangers children's health badly and radioimmunoscintigraphy will provide clue for clinical early diagnosis.We adopted hybridoma technique to develop anti-NB McAb.It was labeled with ^(99m)Tc by a direct pretinning method.Athymic mude mouse bearing human NB were injected with ^(99m)Tc labeled McAb and pooled mouse IgG respectively.ID% (percent injected dose per gram tissue) and T/NT were determined.Radiolabeling efficiency of McAb was 97% and immunoreactivity of McAb after being labeled with ^(99m)Tc was about 85%.The percentage of injected dose (McAb) in tumor increased while other tissue decreased,T/NT ascended and tumor xenografts were clearly visualized as time went on which were in contrast with control group.It is concluded that the developed anti-NB McAb,after being labeled with ^(99m)Tc,can be used to visualize NB xenografts in nude mice.
