摘要
目的研究通脉大生片对排卵障碍型不孕大鼠的促卵泡激素(FSH),促黄体生成素(LH),转化生长因子-β1(TGF-β1)的影响,以探讨通脉大生片治疗排卵障碍不孕症的作用机理。方法建立雄激素致排卵障碍型大鼠模型,随机分为空白对照组,模型组,通脉大生片高剂量组,通脉大生片中剂量组,通脉大生片低剂量组,氯米芬片组。第80日龄开始给药,5周后股动脉取血,用酶联免疫法检测血清中FSH、LH和TGF-β1的含量。取出新鲜的卵巢组织,病理切片HE染色后观察卵巢组织的形态学。结果通脉大生片高剂量组FSH水平高于模型组P<0.05,差异有显著性。通脉大生片高、中、低剂量组LH水平均高于模型组,P<0.01,差异有显著性。通脉大生片高剂量组TGF-β1水平高于模型组,P<0.01,差异有显著性。结论通脉大生片可能通过提高垂体的内分泌功能提高FSH、LH水平;通脉大生片可以升高大鼠血清内TGF-β1水平,其导致该效应的机理,尚需进一步探讨。
Objective To study the influence of Tongmaidasheng-Tablet (TMDST) on follicle- stimulating hormone (FSH), luteinizing hormone (LH) and TGF-β1 of rats with anovulatory infertility, and to explore the mechanism of TMDST for anovulatory infertility. Methods Anovulatory rat models were induced by androgen, and the rats were randomly divided into blank control group, model group, TMDST high-dose group, TMDST medium-dose group, TMDST low-dose group, and clomiphene group. The medi- cation was started at the 80-days-old, then the blood was taken through femoral artery 5 weeks later. The level of serum FSH, LH and TGF-β1 was detected by enzyme-linked immunosorbent assay. Meanwhile, the fresh ovarian tissue was taken out, and morphological examination was observed after HE. Results The level of FSH in TMDST high-dose group is higher than model group, P 〈 0. 05, the difference is signif- icant. The level of LH in All dose of TMDST is higher than model group, P 〈0. 01, the difference is sig- nificant. The level of TGF-β1 in TMDST high-dose group is higher than model group, P 〈 0. 01, the differ- ence was significant. Conclusion TMDST can improve FSH,LH through improving the function of pituita- ry endocrine. TMDST can increase the level of serum TGF-β1 of rats. The further study needs to be ex- plored about its mechanism.
