谷氨酸转运体摄取抑制剂对大鼠脑缺血神经元死亡的影响 被引量:1

摘要 实验观察了谷氨酸转运体摄制剂L-反式吡咯烷-2,4-二羧酸(L-trans-PDC)对脑缺血大鼠神经元死亡的影响。脑室注射L-trans-PDC5μg使大鼠脑梗塞体积显著增大;侧脑室注射L-trans-PDC1μg,2μg或5μg对大鼠脑缺血后脑血流量无显著影响,表明抑制谷氨酸转运体摄取将加重脑缺血致神经元死亡,提示谷氨酸转运体在限制脑缺血时谷氨酸神经毒中起重要作用。 The effect of L-trans-pyrrolidine-2, 4-ducarboxylate(L-trans-PDC), a glutamate uptake inhibitor,on cerebral ischemic neuronal death was investigated.The results were observed as follows: Injection (icv) of L-trans-PDC 1μg or 2μg did not increase significantly infarction volumes (P>0.05). In the group injected(icv) with 5μg L-trans-PDC, infarction volume increased significantly(P<0.05). Administration(icv)with 1μg, 2μg or 5μg L-transPDC did not affect cerebral blood flow in the rats with cerebral ischemia(p>0. 05). The results indicated that inhibition of glutamate transporter uptake aggravated neuronal death induced by cerebral ischemia.It suggests that glutamate transportors may play an important role in limiting neurotoxicity of glutamate under cerebral ischemic conditions.
