Survivin在肾透明细胞癌786-O细胞株中表达的实验研究

Experimental study on the expression of survivin in renal clear cell carcinoma 786-O

目的:检测抗凋亡基因survivin在肾透明细胞癌786-O细胞株中的表达并探讨其临床意义。方法:运用MTT(四唑盐比色)法检测细胞生长状态并绘制生长曲线,运用RT-PCR(逆转录聚合酶链反应)法和免疫组化法检测786-O细胞株中survivin mRNA及survivin蛋白的表达情况。结果:检测出786-O细胞株中有survivin mRNA的表达,survivin蛋白的表达亦呈阳性,而正常肾细胞无阳性表达。结论:survivin mRNA及survivin蛋白在肾透明细胞癌中均有明显的表达,利用这一表达特异性,为以survivin基因为靶点的肾癌基因治疗研究提供了实验依据。

Objective：To investigate the expression and the clinical significance of survivin in renal clear cell carcinoma 786-O. Methods ：The expression of survivin mRNA and protein were detected by RT-PCR and immunohistochemisty,The growth curve of renal clear cell carcinoma 786-O were detected by MTF. Results：The survivin mRNA and protein were detected in 786-O cells.In contrast, no expression of survivin was found in normal renal cells. Conclusion：The survivin gene and protein were significantly expressed in renal clear cell carcinoma 786-O. This may provide the experimental evidence for the survivin gene as a target of gene therapy in renal cancer.
