参附注射液对大鼠心肌缺血再灌注损伤保护作用的实验研究 被引量:12

摘要 目的:探讨参附注射液(Shenfuinjection,SFI)对心肌缺血再灌注损伤(Myocardial ischemia-reperfusion injury,MIRI)的功效。方法:通过复制大鼠心肌缺血再灌注损伤模型,观察SFI不同剂量对心肌缺血再灌注损伤的保护作用。结果:SFI高剂量组对大鼠心肌缺血再灌注损伤模型组在给药后30、60、120min能明显降低大鼠ECG的ST段和T波的偏移,有显著性差异(P<0.05);在给药后120min能明显降低大鼠血清CK、CK-MB、LDH和MMP-9的活性,有显著性差异(P<0.05);还能显著降低大鼠心室脏器系数,使其心肌梗塞面积明显缩小,梗塞区重量减轻,明显改善心肌缺血状况;对冠脉结扎再灌注损伤大鼠全血粘度(低切)和血浆粘度的升高有一定的降低作用。结论:SFI对大鼠心肌缺血再灌注损伤有明显的保护作用,为其临床应用提供了进一步的实验室依据。 Objective :To investigate the function of shenfu injection in myocardial ischemia-reperfusion injury (MIRI). Methods: To observe the effects of Shenfu injection (SFI) in different dosages. An experimental rat' s model of myocardial ischemia- reperfusion inury (MIRI) was created. Results: The high-dosed-Shenfu-injection (SFI) group (20 ml/kg) compared witll the MIRI model group showed significant depressant effect on the ECG ST segment and T wave after 30,60,120 minutes, respectively, of the injection. Such differences were of great statistical value (P〈0.05). Similarly, a reduced serum CK, CK-MB, LDH, MMP-9 activity occurred 120 minutes after the injection (P〈0.05). It could significantly reduce the area and extent of myocardial infarction, obviously improve the situation of myocardial ischemia. It showed a certain reducing effect in the increased whole blood viscosity (low shear) and plasma viscosity. Conclusion:SF] contributing a valid protective effect in myocardial ischemia-reperfusion injury was indeed significant and served as a contributing laboratory basis for further clinical application.
