摘要
目的正交实验优选岩黄连药材的最佳提取工艺。方法以岩黄连中总生物碱、脱氢卡维丁的含量为指标,比较乙醇和酸水两种不同提取溶媒以及回流、渗漉两种不同提取方法,采用L9(34)正交设计,对影响岩黄连提取工艺中的溶剂浓度、溶剂用量、提取时间、提取次数等4个因素进行优化筛选。结果乙醇提取优于酸水提取,回流法提取效果优于渗漉法。醇提最佳工艺条件为:用75%乙醇提取,每次加醇量10倍,提取3次,每次提取时间2h。结论优选得到的工艺简便、稳定、可行。
Objective To optimize the extracting technics for isolating total alkaloids and dehydrocavidin from Corydalis saxicola Bunting by orthogonal design. Methods Corydalis saxicola Bunting was extracted with ethanol or acid by reflux and percolation, respectively. Then the four factors caused the quantity and concentration of solvents, extracting time and frequency were optimized. Results The ethanol refluxing process was better than other processes for isolating total alkaloids and dehydrocavidin from Corydalis saxicola Bunting. The optimal condition was as follows : extracted the crude drug with 10 times more 75% ethanol for 3 times (120 min/time). Conclusion This optimized extraction process is simple, stable and feasible.
