反相高效液相色谱法测定荷叶口服液中荷叶碱的含量

Content Determination of Nuciferine in Folium Nelumbinis Oral Liquid by RP-HPLC

目的建立高效液相色谱法测定荷叶口服液中荷叶碱含量。方法固定相:Hypersil BDS C18柱(4.6mm×150mm,5μm);流动相:甲醇:水:三乙胺:冰醋酸(50:48:1.4:0.7);流速:0.7mL.min-1;检测波长:270nm;柱温:40℃;进样量:10μL。结果荷叶碱在1.282～25.640μg·mL-1范围内线性关系良好(r=0.9991);3份荷叶碱的平均加样回收率分别为99.7%(RSD=1.25%),101.0%(RSD=0.95%),99.5%(RSD=1.14%)。结论该方法准确可靠,快速灵敏,可以作为荷叶口服液中荷叶碱的质量控制标准。

Objective To establish a RP-HPLC method for content determination of nuciferine in Folium nelumbinis oral liquid. Methods Reverse-phase high performance liquid chromatography （RP-HPLC） was carried out with a HypersilBDS C18 column （4.6 mm×150 mm,5 μm） at 40 ℃. Mixture of methanol-water-triethylamide-acetic acid （50 ： 48 ： 1.4 ： 0.7） was used as mobile phase at a flow rate of 0.7 mL·min^-1 with detection at 270 nm. Results Nuciferine showeded a good linear at the range of 1. 282 - 25. 640 μg · mL^-1 （ r = 0. 999 1 ）. The average recovery was 99.7% （ RSD = 1.25% ）, 101.0% （RSD = 0.95% ） and 99.5% （ RSD = 1. 14% ）, respectively. Conclusion The method is accurate, sensitive and can be used for the quality control of Folium nelumbinis oral liquid.
