摘要
以3种牡丹的茎尖为外植体,研究不同品种牡丹茎尖在不同激素配比条件下,愈伤组织和丛生芽的诱导、芽的生长及生根情况。结果表明:适合3种牡丹丛生芽产生的最佳培养基,紫斑牡丹为MS+6-BA 0.5 mg/L+IBA 0.2 mg/L+GA30.1 mg/L+Vc 100 mg/L,青心白为MS+6-BA 0.5 mg/L+IBA 0.5 mg/L+CH 100 mg/L,朝霞为MS+6-BA 1.0 mg/L+IBA 0.5mg/L+CH 100 mg/L+Vc 100 mg/L;适合3种牡丹生根的培养基为1/2MS+IAA 1.0 mg/L+IBA 2.0 mg/L+NAA 0.2 mg/L,其中,朝霞生根效果最佳,生根率可达82.35%,紫斑牡丹和青心白分别为75%和50%;活性炭和Vc可有效防止褐化的发生。
The stem apex of three peony were used to research induction of callus and cluster buds,buds growth and rooting in the compounding proportions of different hormones. The results indicated that the optimal medium for cluster buds inducing of three peony, Paeonia rockii was MS+6-BA 0. 5 mg/L+IBA 0. 2 mg/L+GA3 0. 1 mg/L+Vc 100 mg/L,cv. QingXinBai was MS+6-BA 0. 5 mg/L+IBA 0. 5 mg/L+CH 100 mg/L, cv. Zhao Xia was Ms+6-BA 1.0 mg/L+IBA 0. 5 mg/L+CH 100 mg/L+Vc 100 mg/L;the optimal medium for rooting was 1/2MS+IAA 1.0 mg/L+IBA 2. 0 mg/L+NAA 0. 2 mg/L, the rooting rate was 82. 35G of cv. ZhaoXia and the best,Paeonia rockii and cv. QingXinBai were 75% and 50% ;activated carbon and Vc could prevent the occurrence of browning effectively.
