摘要
为解决赤灵芝生产中多次无性繁殖而常出现菌种老化和退化问题,优选适合赤灵芝母种的培养剂。利用四因素五水平二次通用旋转组合设计,研究葡萄糖、蛋白胨、硫酸镁、磷酸二氢钾对赤灵芝菌丝的长速、干重的影响。结果表明:葡萄糖、蛋白胨、酵母粉、无机盐(MgSO4·KH2PO4)4因素中,对赤灵芝菌丝长速的效应为葡萄糖〉蛋白胨〉酵母粉〉无机盐;对菌丝干重的效应为葡萄糖〉酵母粉〉无机盐〉蛋白胨。获得赤灵芝菌丝的长速〉1.31cm/d,菌丝干重〉26.09mg/tube,用葡萄糖16.37~17.05g/L,蛋白胨1.617~1.689g/L、酵母粉1.872~2.056g/L、无机盐(MgSO4·KH2PO4)2.352~2.706g/L。
