摘要 目的检测尿路上皮癌组织中CD146表达和微血管密度(MVD),并探讨其意义。方法采用实时荧光定量PCR技术和免疫组化法检测尿路上皮癌组织及正常尿路上皮组织中CD146mRNA及其蛋白的表达情况,用CD34标记免疫组化法检测MVD。结果癌组织中CD146mRNA、蛋白表达及MVD均明显高于正常黏膜组织,P均<0.05。癌组织中CD146mRNA、蛋白表达与肿瘤病理分级、临床分期、淋巴结转移状况及MVD有相关性,P均<0.05。结论尿路上皮癌组织中CD146表达和MVD升高。其尿路上皮癌的分化、浸润及淋巴转移有关。CD146可能通过促进肿瘤血管生成在尿路上皮癌发生发展过程中发挥重要作用。 Objective To observe the change of CD146 expression and microvessel density in urothelial cancer tissue. Method Real time polymerase chain reaction (RT-PCR) and immunohistochemisty was used to detect the expression of CD146 and microvessel density (MVD) in urothelial cancer tissues and normal urothelial tissues. Result The quantity of CD146 mRNA in urothelial cancer was higher than that in normal Urothelial tissues (P 〈 0.01 ). Moreover,the expression in creases gradually with the grades going up (P 〈 0.01 ). With the infiltration deepening and appearance of lymph node me tastasis, the expression of CD146 gene mRNA increased ( P 〈 0.01 ). The expression of CD146 protein was significant higher in tumor tissue than those in normal tissue( P 〈 0.01 ). The expression of CD146 in Urothelial Cancer with the grades going up, CD146 became higher gradually( P 〈 0.01 ). The expression of CD146 protein in deeper invasive group and group with lymph node metastasis was significant higher than those in superficial invasive group and group without lymph node metastasis (P 〈 0.05 ). The MVD in urothelial cancer tissues was higher than that in normal urothelial tissues (P 〈 0.01 ). MVD in positive expression of CD146 in urothelial cancer tissues were higher than those in the negative urothelial cancer tissues ( P 〈 0. 05 ). Conclusion The expression of CD146 and MVD increase in urothelial cancer tissues. It may be related to the differen tiation, and progression and metastasis of urothelial cancer. CD146 may play important roles in angiogenesis of urothelial cancer.
