抗纤复方Ⅰ号对酒精性肝病大鼠纤溶酶原激活物抑制剂-1的影响 被引量:1

摘要 目的探讨纤溶酶原激活物抑制剂-1(PAI-1)在酒精性肝病大鼠肝脏中的表达及抗纤复方Ⅰ号(KXⅠ)对其表达的影响。方法Wistar大鼠给予乙醇8~10g.kg-1.d-1,3次灌胃,共12周,制备酒精性肝病大鼠模型为酒精组,对照组给予生理盐水2.0mL/只,KXⅠ组在给予同酒精组等量乙醇的基础上,给予KXⅠ号水煎剂4.0mL.kg-1.d-1,2次灌胃,共12周。实验4、8、12周末分批处正常组、酒精组、中药组大鼠,苏木素-伊红(HE)染色观察肝脏组织学改变。应用免疫组织化学技术检测PAI-1在酒精性肝病过程中的定位及表达,并用病理图像分析仪对其结果进行半定量分析。结果12周末,肝小叶脂肪变,肝细胞坏死增多,炎性细胞浸润明显;中药组肝细胞坏死及炎细胞浸润不明显。PAI-1在正常肝脏组织中央静脉区细胞的胞质中基本不表达,酒精组:4周末开始,以中央静脉为中心,肝脏细胞胞质中PAI-1表达增多,且随着酒精刺激时间的延长,肝脏细胞胞质中PAI-1的表达呈进行性增加,差异有统计学意义(P<0.01)。结论酒精性肝病过程中,PAI-1表达进行性增强,KXⅠ号能抑制PAI-1的表达,延缓肝纤维化的形成和发展。 Objective To investigate the expression of plasminogen activator inhibitor type Ⅰ(PAI-1) in rats with alcoholic liver disease and the effect of Kangxian Fufang Ⅰ (KX Ⅰ ) on PAI-1 expression. Methods Wistar rats weighing 160 to 190 g were lavaged with normal saline (2.0 mL) ,ethanol (8-10 g · kg^-1 · d^-1 ) ,and ethanol plus KX Ⅰ (4.0 mL · kg^-1 · d^-1 ) for 12 weeks in control group, alcohol group and KX Ⅰ group , respectively. At the end of 4,8 and 12 weeks, the rats were killed, and the histopathological changes of the liver were observed. The PAI-1 expression in the liver was examined by immunohistochemical method. Results At the end of 12 weeks, steatosis, inflammation and necrosis were observed in alcohol group. In contrast, these alterations were attenuated by KX Ⅰ administration. In normal liver, PAI-1 expressed in the cell plasma of liver, whereas the expression of PAI-1 was located around central vein from 4 weeks. Conclusion In rats with alcoholic liver disease, PAI-1 is highly expressed, and KX Ⅰ could decrease PAI-1 expression.
