摘要
本文研究了梨头霉菌催化11-脱氧皮质醇醋酸酯(RSA)转化氢化可的松过程中,有机相中底物浓度、转化环境、菌体成熟状态以及有机相所占比例等因素对转化反应的影响.实验结果表明,收集pH3.8~4.1的发酵液,在12.5%的丙二醇转化体系中,将底物浓度由2.8‰提高到4.5‰,底物仍能被完全转化且目的产物(β-异构体)的生成与低浓度时相近.
In order to raise the yield of hydrocortisone from RSA, the conversion by Absidia orchidis was conducted in a propandiol system. The effects of substrate concentration, state of thallus maturation, propandiol concentration and the pH value of conversion were studied when transformed to hydrocortisone by Absidia orchidis. Experimental results showed when the RSA concentration was raised 75% the substrate is still transformed completely in 12.5% propandiol system. The optimum pH value is controlled at 3.8 ̄4.1 on culture thallus, and the pH 5.8 ̄6.2 is more advantageous to transform in propantiol system by Absidia orchidis.
