昆明山海棠对中国仓鼠V79细胞核/质分裂协调性的影响 被引量:1

摘要 本研究以哺乳动物非整倍体诱发剂昆明山海棠根部水抽提物(TripterygiumHypoglau-cum(Level)Hutch,THH)处理中国仓鼠V97细胞,通过检测V79细胞末期和早G1期核/质构型变化和双核间期细胞的频率,分析了THH对哺乳动物细胞质分裂的影响。结果指出:THH能显著诱发V79细胞的异常质裂(P<0.001),并明显的提高双核细胞的频率(P<0.001-0.05)。结合以往的工作,提示THH不仅对哺乳动物染色体正常分离有直接的影响,同时对细胞质的正常分裂也具有显著抑制作用。受试物可能通过多途径诱发哺乳动物非整倍体。 V79 Chinese hamster cells were treated in vitro by Tripterygium Hypoglaucum (Level) Hutch(THH), Which contains some aneugen. The disturbance of mitotic coordination by THH were determined by examining nucleolus/cytoplasmic configuration of cells in late telophase-very early G 1 and the frequency of binuclear cells in interphase cells. The experiment showed that the compound influenced mitotic coordination and significantly increased the frequency of binuclear cells. The result indicated that THH contains some component which inhibits cytokinesis during mammal cell mitosis. According to the research before, we may conclude that THH not only directly induces chromosome non-disjunction but also shares another mechenisms of aneuploid induction related to cytokinesis in mammalian cells.
