摘要 目的制备替莫唑胺脂质体.方法以磷脂和胆固醇为载体制备替莫唑胺脂质体,高速离心法分离脂质体与未包封药物,紫外分光光度法测定脂质体包封率;以包封率为主要评价指标,考察制备方法和制备工艺对替莫唑胺脂质体的影响.结果采用逆相蒸发法制备替莫唑胺脂质体,包封率为33.6±3.8%.结论由于替莫唑胺的溶解性等问题所限,很难提高替莫唑胺脂质体的包封率. OBJECTIVE To prepare temozolomide liposomes. METHODS The temozolomide liposomes were prepared with phospholipid and cholesterol. To separate the liposomes and free drugs with high speed centrifugation. The entrapment efficiency was determined by ultraviolet spectrophotometry. To investigate the effects to the temozolomide liposomes of the prepration methods and technology on the basis of entrapment efficiency. RESULTS The entrapment efficiency of temozolomide liposomes prepared by the antiphase evaporation method was (33.6±3.8) %. CONCLUSION It is hard to raise the entrapment efficiency of temozolomide liposomes because of the limitation of the temozolomlde's dissolvability.
