摘要
目的探讨小鼠部分胰腺切除手术诱导的糖尿病以及对胰岛细胞再生的影响,以建立研究胰岛细胞再生的动物模型。方法对3月龄近交品系C57B6小鼠胰腺切除65%~70%,通过代谢方法、糖耐量和免疫组织化学的方法,研究该方法对血糖和胰岛细胞的再生影响,以确定该模型是否能用于糖尿病的研究。结果小鼠胰腺部分切除,术后一周内小鼠糖耐量受损。术后第14天和第28天的糖耐量实验结果表明3月龄小鼠的糖耐量随着时间的推移而逐渐恢复。免疫组化结果显示3月龄小鼠术后1周胰岛β细胞再生率为(15.1±10.0)%,2周为(25.4±8.1)%,4周为(45.7±14.6)%。结论胰腺部分切除手术能够一定程度上诱导小鼠产生糖尿病,并且诱导胰岛β细胞的再生,从而可用于研究胰岛β细胞再生的基础研究。
Objective To study the effect of mice partial pancreatectomy on islet regeneration and diabetes,and to evaluate this partial pancreatectomy model in research on islet regeneration.Methods 3-month-old inbred C57B6 mice were used in this 65%70% partial pancreatectomy operation.Glucose metabolism method,glucose tolerance test and immunohistochemistry Methods were used to evaluate the changes in blood glucose and the ability of islet regeneration.Thus drew the conclusion whether this model could be used in diabetes research.Results One week after partial pancreatectomy,mice had impaired glucose tolerance.In IVGTT data of the following 14 days and 28 days,3-month-old mice gradually recovered from this injury.Immunohistochemistry staining Results showed the ability of islet beta cell regeneration was (15.1±10.0)% in one week,(25.4±8.1)% in 2 weeks and (45.7±14.6)% in 4 weeks respectively. Conclusion Partial pancreatectomy could induce mice diabetes in some degree,and thus induce islet beta cell regeneration.So this model could be used in basic research on islet regeneration.
