安宫牛黄丸药效组分对内毒素损伤小鼠脑组织一氧化氮和一氧化氮合酶的影响 被引量:7

Influence of Effective Constituents of Angongniuhuang Pill on Nitric Oxide and Nitric Oxide Syntheses of Endotoxin Injury Mice’s Brain Tissue

摘要 目的:观察安宫牛黄丸药效组分对内毒素损伤小鼠脑组织一氧化氮(nitric oxide,NO)系统的影响,探讨安宫牛黄丸药效组分与安宫牛黄丸的等效性、中药药效组分理论的可行性及其阻抑内毒素致脑损伤作用的可能的机制。方法:以小鼠腹腔注射脂多糖建立内毒素损伤模型;HE染色观察脑组织病理损伤情况;生化法检测脑组织中NO和一氧化氮合酶(nitric oxide synthase,NOS)含量。结果:内毒素损伤小鼠脑组织出现以神经细胞肿胀、核固缩、胶质细胞增生、淋巴细胞浸润为特征的病理变化;NO含量明显增多,总一氧化氮合酶(T-NOS)和诱导型一氧化氮(inducible nitric oxide synthase,iNOS)活力显著增强。安宫牛黄丸及其药效组分各剂量组小鼠脑组织病理变化减轻,神经细胞肿胀、核固缩、胶质细胞增生等病变有所好转;NO含量、T-NOS、iNOS活力均有不同程度降低。结论:安宫牛黄丸药效组分对脑损伤有保护作用,提示该药效组分与安宫牛黄丸的开窍、醒神功效具有一致性。其可能通过阻抑iNOS诱导产生过量的NO,产生神经保护作用。 Objective: To observe the influence of effective constituents of Angongniuhuang Pill on nitric oxide system in endotoxin injury mice's brain tissue, investigate the efficacy Angongniuhuang Pill component equivalence and Angongniuhuang Pill, and efficacy of Chinese medicine theory component of the feasibility; explore the possible molecular mechanism of its inhibition on the brain damage caused by endotoxin. Methods: Endotoxin injury model was founded by intraperitoneal injecting LPS into mice. Pathological situation was observed through HIM staining; content of nitric oxide(NO)and nitric oxide syntheses(NOS)in brain tissue were measured by biochemical tests. Results: Pathological changes in endotoxin injury mice's brain tissue occurred featured by neuron swelling, karyopyknosis, glial cells hyperplasia and lymphocyte infiltration. Content of NO, activity of T-NOS and inducible nitric oxide syntheses(iNOS) in brain tissue increased significantly. Pathological changes of Angongniuhuang Pill group and its effective constituents groups with different dose were alleviated, and content of NO, activity of T-NOS and iNOS also decreased. Conclusion: Components of efficacy Angongniuhuang Pill has a protective effect on brain injury, suggesting that the effective component of the effectiveness and consistency Angongniuhuang Pill. All results showed that neural protective effect of effective constituents of Angongniuhuang Pill probably appeared through inhibiting iNOS on inducing excessive NO.
