吡格列酮对兔动脉粥样硬化血管壁ICAM-1表达的影响 被引量:3

摘要 目的:探讨吡格列酮对动脉粥样硬化(Atherosclerosis,AS)血管壁细胞间粘附分子(Intercellular adhesion molecule-1,ICAM-1)表达的影响。方法:新西兰兔随机分成5组:A:正常对照,B:正常+低剂量吡格列酮(Pioglitazone,PIO),C:病理对照,D:病理+低剂量PIO,E:病理+高剂量PIO。采用免疫组化及逆转录聚合酶链反应(RT-PCR)的方法观察吡格列酮血管壁ICAM-1蛋白及mRNA表达的影响。结果:AS斑块内有ICAM-1蛋白和mRNA高表达,PIO可抑制ICAM-1蛋白和mRNA表达(P<0.05),高剂量较低剂量抑制程度更大(P<0.05)。结论:PIO可抑制AS斑块ICAM-1的mRNA和蛋白表达,进而发挥抗AS炎症的作用。 Objective:To explore the effect of pioglitazone (PIO) on expression of intercellular adhesion molecule-1 (ICAM-1) in atherosclerotic rabbit aorta. Methods : Newzealand white rabbits were divide into 5 groups : normal control, normal+low dose of PIO, and AS control,AS+low dose of PIO,AS+high dose of PiO. Immunohistochemistry was used to detect the protein of ICAM-1. Reverse transcription -polymerase chain reaction (RT-PCR) was used to detect the mRNA of ICAM-1. Results:ICAM-1 protein and mRNA in AS plaques. PIO could inhibit the expression of ICAM- 1 protein and mRNA (p〈0.05). The effect of high dose was more than that of were highly expressed low dose(P〈0.05 ). Conclusion: PIO could inhibit the expression of ICAM-1 and further produce its anti-inflammatory effect on AS.
