CDX2和COX2在人大肠肿瘤中的表达特点及与转移的关系 被引量:6

Expression of Caudal-Relatedhomeodomain Transcription 2 and Cyclooxygenase-2 in Human Colorectal Neoplasms and its Relation to Metastasis

摘要 目的研究尾型同源盒转录因子2(CDX2)和环氧合酶2(COX2)在人大肠腺癌发生发展过程中的作用,并探讨其与大肠腺癌侵袭和转移的关系.方法采用免疫组化技术SP法,观察在人大肠腺瘤30例、无淋巴结转移的大肠腺癌35例、有淋巴结转移的大肠腺癌35例组织中CDX2、COX2蛋白的表达特点.结果COX2蛋白定位于细胞浆和细胞膜,从大肠腺瘤到大肠腺癌其蛋白表达明显增强(P=0.000);从高分化的大肠腺癌到低分化的大肠腺癌之间其蛋白表达明显增强(P=0.009);在无淋巴结转移的大肠腺癌和有淋巴结转移的大肠腺癌原发灶之间其蛋白表达差异有统计学意义,且呈正相关(P=0.035,rs=0.253).CDX2蛋白定位于细胞核,从大肠腺瘤到大肠腺癌其蛋白表达明显降低(P=0.000);从高分化的大肠腺癌到低分化的大肠腺癌之间其蛋白表达也明显降低(P=0.038);在无淋巴结转移的大肠腺癌和有淋巴结转移的大肠腺癌原发灶之间其蛋白表达差异有统计学意义,且呈负相关(P=0.048,rs=-0.237).CDX2蛋白和COX2蛋白的表达在大肠腺癌中呈负相关(P=0.028,rs=-0.220).结论COX2和CDX2可作为判断大肠腺癌患者预后的指标.CDX2蛋白与COX2蛋白在大肠腺癌中的作用有负相关关系. Objective To investigate roles of caudal-relatedhomeodomain transcription-2 (CDX2) and Cyclooxygenase-2 (COX2) protein in the carcinogenesis, progression, invasion and metastasis of human colorectal adenocarcinomas and identify its relation with the clinicopathological indices of colorectal adenocarcinoma. Methods CDX2 and COX2 protein were detected by SP-immunohistochemistry in formalin-fixed and paraffin-embeded tissues of 30 colorectal adenoma tissues, 35 colorectal adenocarcinoma tissues with no regional lymph nodes metastasis, 35 primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis. Results The expression of COX2 protein distributed in cytoplasms and cell membranes, it was gradually increased from colorectal adenoma tissues to colorectal adenocarcinoma tissues (P = 0.000). From the well differentiated colorectal adenocarcinoma tissues to the poor differentiated tissues the expression of COX2 protein was significantly increased (P =0.009). And the expression of COX2 protein between colorectal adenocarcinoma tissues with no regional lymph nodes metastasis and primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis had significant difference (P = 0.035, rs = 0.253). The expressions of CDX2 protein distributed in karyon, it was gradually decreased from colorectal adenoma tissues to colorectal adenocarcinoma tissues (P = 0.000). From the well differentiated colorectal adenocarcinoma tissues to the poor differentiated tissues the expression of CDX2 protein was significantly decreased (P = 0.038). And the expression of CDX2 protein between colorectal adenocarcinoma tissues with no regional lymph nodes metastasis and primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis had significant difference (P = 0.048, rs =-0.237). Conclusion The down-regulation of CDX2 protein and the up-regulation of COX2 may play important roles in colorectal carcinogenesis and progression.
