黄芩苷对脑缺血-再灌注小鼠纹状体Bax和Bcl-2蛋白表达影响 被引量:4

摘要 目的探讨黄芩苷对脑缺血-再灌注损伤小鼠脑纹状体神经细胞的Bax/Bcl-2表达水平的影响.方法昆明小鼠30只,随机分为假手术组(n=10)、再灌注组(n=10)及黄芩苷组(n=10).夹闭小鼠颈总动脉方式建立脑缺血模型,免疫蛋白印记法检测Bax和Bcl-2表达及其比值变化.结果在脑缺血-再灌注损伤4d后,Bcl-2与Bax表达都上调,黄芩苷预处理可以下调小鼠脑组织纹状体区神经细胞Bax的表达水平,降低Bax/Bcl-2比值(P<0.05).结论黄芩苷能够下调Bax/Bcl-2的比值,可能对脑缺血-再灌注损伤具有抗神经元凋亡保护的作用. Objective To explore the effect of baicalin on Bax and Bcl-2 expression in mouse model of cerbral isehemia-reperfusion. Methods Thirty Kunming mice were randomly divided into control group (n = 10) , reperfusion group (n = 10) and baicalin group (n = 10). The mouse ischemia-reperfusion injury model was established by carotid artery clamping. Intraperitoneal Baicalin injection was administered 14 days before ischemia-reperfusion injury model establishment. Western blot analysis was used to detect Bax and Bcl-2 protein expression. Results The expression of Bel-2 and Bax was upregnlated 4 d after cerebral ehemia-reperfusion injury. Baicalin treatment significantly reduced the expression of Bax and Bcl-2 after ischemia-reperfusion injury, and Bax/Bel-2 ratio was reduced (P 〈 0.05). Conclusion Baiealin has protective effects on neuron against ischemia-reperfusion injury, and the mechanism may be related to downregnlating Bax/Bcl-2 ratio.
