白介素12联合组织途径抑制因子2基因转染对喉癌细胞侵袭与凋亡的影响 被引量:1

Effection of IL-12 and TFPI-2 transfected in invasion and apoptosis of laryngeal squamous carcinoma

摘要 目的探讨腺病毒介导的白介素12(IL-12)与组织途径抑制因子2(TFPI-2)基因联合转染对喉鳞癌细胞侵袭及凋亡的影响。方法将IL-12与TFPI-2体外转染喉鳞癌Hep-2细胞,酶联免疫吸附(ELISA)法检测IL-12和TFPI-2蛋白在Hep-2细胞的表达,应用四甲基偶氨唑盐酶反应比色法检测转染后的喉癌细胞生长与增殖情况,Boyden侵袭小室实验检测Hep-2细胞侵袭能力的变化,应用流式细胞仪检测喉癌细胞的凋亡。结果ELISA结果显示IL-12和TFPI-2在实验组Hep-2细胞表达增高,Boyden侵袭小室法显示实验组Hep-2细胞的侵袭能力降低,流式细胞仪显示实验组Hep-2细胞凋亡显著增加。结论IL—12与TFPI-2基因联合转染能有效抑制人喉鳞癌Hep-2细胞的馒袭能力并促进其凋亡。 Objective To study the effects of adenovirus-mediated IL-12 and TFPI-2 gene in the invasion and apoptosis of laryngeal squamous cancer. Methods IL-12 and TFPI-2 were transfected into laryngeal squamous cancer (Hep-2) cell. ELISA was used to test IL-12 and TFPI-2 protein expression and MTT was used to examine the proliferative ability of Ad-IL-12 and Ad-TFPI-2 transfected Hep-2 cells. Furthermore, Boyden Chamber and flow cytometry were used to test the invasive ability and apoptosis of Ad-IL-12 and Ad-TFPI-2 transfected Hep-2 cells. Resuits In the Hep-2 cells of treated group,the IL-12 and TFPI-2 expression were increased while the proliferative capability was descent compared with the control. Boyden Chamber showed the invasive ability was significantly decreased and flow cytometry showed the apoptosis was obviously increased in the Ad-IL-12 and Ad-TFPI-2 transfected Hep-2 ceils. Conclusion IL-12 and TFPI-2 recombinant adenovirus can effectively inhibit the invasive capability and promote apoptosis of laryngeal squamous cancer.
