摘要
目的评价受体血清保存7 d的兔角膜行角膜移植术后植片的内皮细胞功能。方法30只新西兰大白兔随机分为2组:A组(对照组)15只和B组(实验组)15只。以传统湿房保存1 d的兔角膜作为A组供体;以受体血清保存7 d的兔角膜作为B组供体,对2组新西兰大白兔行角膜移植术。裂隙灯观察2组术后第3天,第1、8周角膜植片(大体形态)情况。2组术后第8周取角膜植片行扫描电镜观察超微结构及锥虫蓝-茜素红检测角膜内皮细胞活性;角膜厚度仪测量角膜厚度及图像分析系统计算内皮细胞密度和细胞面积。结果A、B 2组术后第8周眼部充血消退,植片透明、清亮,未见排斥反应。A组细胞密度、细胞面积与B组比较,差异均有统计学意义(均P<0.01),A组角膜厚度与B组比较,差异无统计学意义(P>0.05)。结论受体血清有维护角膜内皮细胞结构和功能的作用。
Objective To evaluate the postoperative graft and function of rabbit's endothelial cell on the corneal preservation in recipient's serum for 7 days by corneal transplantation.MethodsThirty New Zealand big white rabbit were randomly divided into 2 groups:15 only in group A(control group) and 15 only in group B(experimental group).The rabbit corneal preservation with the traditional moist chamber for 1 days as the group A donor,preservation with recipient serum for 7 days as the group B donor,penetrating corneal transplantation with the rabbit of both groups.At the 3th day,the 1th week,the 8th week postoperative corneal graft,the corneal grafts were observed(gross morphologic) by slit-lamp of 2 groups.At the 8th week,those were observed for ultra-microstructure by scanning electron microscope and the corneal endothelial cells activity were detectd by the trpan blue-alizarin red stain;the corneal thickness was measure;corneal endothelial cells density were calculated and cells site were observed by the Image analysis system.Results At 8th weeks after experiments conjunctival congestion was reduced in both groups.Graft transparent was clear and there was no rejection.In group B,after corneal transplantation 2 months the cell density and cell size were statically significant different compared with that in group A(all P0.01).However there was no significant difference of corneal thickness between two groups(P0.05).Conclusion Recipient serum has the function with supporting the structure and function of rabbit's corneal endothelial cell.
