摘要
为建立曼地亚红豆杉中紫杉醇含量测定的反向高效液相色谱分析方法,采用Diamond C18(250mm×4.6mm,5μm)色谱柱,柱温30℃;乙腈-水(52:48)作为流动相,流速1.0mL/min;检测波长227/nm。经测定曼地亚红豆杉枝叶中紫杉醇平均含量达到0.0237%,使用该检测方法可将紫杉醇与其类似物分离。曼地亚红豆杉枝叶中紫杉醇含量较高,该检测方法稳定,可用于紫杉醇的快速定量测定。
To establish a RP-HPLC for the determination of paelitaxel in Taxus media,the samples were separated on a Diamond C18(4.6 mm × 250 mm,5 μm) column, the column was kept at 30 ℃, with the mobile phase consisting of acetonitrile and water(52:48 ), at a flow rate of 1.0 mL/min, the elution was monitored at the wavelength of 227 nm. The average determination of paclitaxel to reach of 0.0237%, chromatographic separation of paclitaxel and its analogues achieved baseline resolution. Paclitaxel with a high level can be fast detection to quantity by the methods.
