摘要
目的观察理气活血法对萎缩性胃炎癌前病变大鼠胃黏膜组织基因表达谱的影响。方法使用金属弹簧、灌服盐水溶液和热糊的综合方法建立大鼠萎缩性胃炎癌前病变模型,运用基因芯片技术,检测理气活血法干预后萎缩性胃炎癌前病变大鼠基因的差异表达变化,筛选出差异表达基因,并应用Real-time PCR对主要差异基因进行鉴定。结果共筛选出差异表达基因14个,其中上调基因8条,下调基因6条,共同构成了理气活血法干预大鼠萎缩性胃炎癌前病变的差异表达基因谱。其中蛋白酪氨酸激酶2(ptk2)在自然恢复组中明显下调,经理气活血法干预后明显上调。结论理气活血法可上调ptk2基因的表达,从而调节肌动蛋白细胞骨架的重构,促进损伤的胃黏膜修复和愈合,这可能是理气活血法治疗萎缩性胃炎癌前病变的机理之一。
Objective To observe the influence of TCM therapy of regulating qi and activating blood circulation on the gene expression profile of gastric mucosa in rats with precancerosis of atrophic gastritis. Methods The rat model of precancerosis of atrophic gastritis was established by the synthetic method of metal spring insertion and feeding of NaCI solution and heat paste.The changes of differential gene expressions in the rats were detected by using gene chip technology after the intervention of the TCM therapy.The differential expression genes were screened and then identified by applying RT-PCR. Results There were totally 14 differential expression genes screened,and among them 8 were up-regulated genes and 6 were down-regulated genes.All of them consisted of the differential expression gene profile after the intervention of the TCM therapy in rat with precancerosis of atrophic gastritis,among which ptk2 was regulated significantly downward in the spontaneous recovery group and regulated significantly upward after the intervention of the TCM therapy.Conclusion The TCM therapy of regulating qi and activating blood circulation can regulate upward the gene expression of ptk2 in order to regulate the remodeling of actin cytoskeleton,and improve the repairing and healing of gastric mucosa.It is may be one of the mechanisms of the TCM therapy for treating precancerosis of atrophic gastritis.
