摘要
以水稻1103s和亲本8902s为材料,研究在降温条件下植株间断失绿性状表达过程中叶绿体蛋白质谱带、蛋白质含量与蛋白酶活性、光合作用变化。结果表明,在降温条件下,随性状表达1103s的叶绿素含量下降,叶绿素a/b值上升;在叶绿体蛋白质谱带中,20KD、25KD、50KD等主带含量减少。从同一叶片性状表达的绿区与黄区叶绿体蛋白质比较看,黄区缺少18KD,这说明同一叶片不同区域叶绿体蛋白质低温反应不同,从而造成蛋白质降解也不同,这是间断失绿性状表达的基础。从降温结束到性状表达(约2d~3d),叶片蛋白质含量下降与蛋白酶活性提高呈显著负相关;当性状逐渐消失时,蛋白质含量与叶绿素含量同步增长。据此认为,1103s的间断失绿性状可能是由核基因控制,该基因在降温条件诱导下启动。
s is a temperature sensitive chlorophyll mutant which has the characteristics of character chequered with green and chlorosis. After the low temperature treatment and the appearing of the character chequered with green and chlorosis in 1103s,the content of chlorophyll decreased and the ratio of chlorophyll a/b increased;the SDS PAGE results indicated the contents of proteins of chloroplast with 20KD,25KD,50KD were lower than those in normal leaves,and 18KD disappeared in chloroplast in the yellow region of the leaves in 1103s.It was an important factor that there are differences between the photosynthesis of chlorophyll in the yellow region and that in the green region.Basing on the change of chloropyll and protein of chloroplast,it is suggested that there be a regulating effect in nucleic gene of the mutant line 1103s during the character appearing period.
