摘要
目的:研究脾虚胃癌转移鼠模型的分子生物学特征;参术胶囊治疗脾虚胃癌转移鼠模型的基因表达谱变化,分析其治疗脾虚胃癌的分子机理。方法:在将BALB/c-nu/nu小鼠采用食醋法建立脾气虚证动物模型的基础上用外科原位移植方法,建立脾虚胃癌转移鼠病证结合动物模型;用基因芯片分别检测脾虚胃癌转移鼠模型组和胃癌细胞株,脾虚胃癌转移鼠模型组和空白对照组,参术胶囊治疗组和模型组,阳性药物的基因差异表达。结果:脾虚胃癌转移鼠模型中95条基因差异表达与代谢、免疫、离子通道和运输蛋白等有关;参术胶囊能逆向调节因脾虚胃癌引起的9条基因(代谢相关基因、离子通道和运输蛋白)的差异表达。结论:脾虚因素加重了脾虚胃癌的病情;参术胶囊是从多靶点出发来治疗脾虚胃癌和改善症状的。
Objective : To study the molecular biological characteristics in the spleen - deficiency gastric cancerometastasis mice model; to study gene expression profile induced by treatment of Shenzhu Capsules on the model with spleen - deficiency and gastric cancerometastasis in mice and to analyze its molecule mechanism of therapeutics on spleen - deficiency gastric cancer. Methods: On the basis of establishment of spleen - qi deficiency models by feeding BALB/c - nu/nu mice with vinegar, an animal model with combination of disease and syndrome called model mice with spleen -deficiency and gastric cancerometastasis was established by adopting SOI to transplant SGC9701 gastric cancer tissure after generation cultivation on the tunica serosa gastria of the mice. Gene chips were used to detect differential expression of the spleen - deficiency gastric cancerometastasis mice and the gastric cancer cell line, the spleen - deficiency gastric cancerometastasis mice and the normal mice, the Weifuchun treated mice and the model mice as well as Shenzhu capsule treated mice and model mice. Results: In the spleen -deficiency gastric cancerometastasis mice model, differential expression of 95 genes was related to spleen- deficiency and attributed to metabolism, immunity, ironic channel and transport proteins, cytoskeleton and cell movement, etc... Shenzhu capsules could retroregulate differential expression of 9 genes (metabolism related genes, ion channel and transport protein, etc. ) caused by spleen -deficiency gastric cancer. Conclusions: Spleen deficiency aggravated the state of gastric cancer. Shenzhu capsules treated spleen deficiency gastric cancer and improved the body by regulating expression of genes.
