食管鳞癌组织中EphrinB2、半胱氨酸蛋白酶8蛋白和mRNA的表达 被引量:3

摘要 目的:探讨人食管鳞癌组织中EphrinB2与半胱氨酸蛋白酶8(Caspase-8)的表达。方法:运用实时荧光定量PCR和免疫组化方法检测96例食管鳞癌患者的癌组织和癌旁正常组织中EphrinB2与Caspase-8的表达,采用Cox回归进行食管癌患者生存分析。结果:癌组织中Caspase-8mRNA(ΔCt)与蛋白的表达水平分别为(7.38±2.47)和(2.51±2.38)%,低于配对正常组织的(5.46±1.87)和(8.85±7.88)%(t=1.375和-22.761,P均<0.001);癌组织中EphrinB2蛋白的表达水平为(11.67±2.48)%,高于配对正常组织中的(1.71±0.60)%(t=-38.322,P<0.001)。癌组织中EphrinB2与Caspase-8蛋白的表达水平相关(r=-0.340,P<0.001)。COX回归分析:有家族史和Caspase-8mRNA低表达是影响食管癌患者生存的危险因素(95%CI分别为1.990~8.547和0.152~0.788,P均<0.05)。结论:人食管鳞癌组织中EphrinB2的高表达和Caspase-8的低表达可作为食管癌发生发展的评估指标。 Aim:To investigate EphrinB2 and Caspase-8 expression in esophageal squamous cell carcinoma tissue.Methods:The expressions of EphrinB2 and Caspase-8 in 96 cases of esophageal squamous cell carcinoma tissues and matched normal esophageal tissues were quantified using real-time PCR assay and immunohistochemistry staining.Cox regression survival analysis was used to estimate the effects of the clinical characteristics.Results:The expression levels of Caspase-8 mRNA and its protein were(7.38±2.47) and(2.51±2.38)%,significantly lower than those(5.46±1.87) and(8.85±7.88)% in their matched normal esophageal tissues(t=1.375 and-22.761,P0.001).The expression levels of EphrinB2 protein was(11.67±2.48)%,significantly higher than that(1.71±0.60)% in matched normal esophageal tissues(t=-38.322,P0.001).The protein expression levels between EphrinB2 and Caspase-8 were correlated(r=-0.340,P0.001).Cox regression survival analysis showed that family history and down-regulation of Caspase-8 mRNA were risk factor for survival of esophageal cancer patients(95% CI were 1.990~8.547 and 0.152~0.788,P0.05).Conclusion:Upregulation of EphrinB2 and downregulation of Caspase-8 may serve as valuable target for esophageal cancer.
