高脂肥胖大鼠肝脏组织中叉头状转录因子O1的表达及其与糖脂代谢的关系 被引量:6

Relationship between FoxO1 expression in liver and glycolipid metabolism in high-fat diet obese rats

摘要 目的:观察高脂肥胖大鼠肝脏组织中叉头状因子O1(FoxO1)的表达,探讨其与血糖、血脂代谢的关系。方法:雄性SD大鼠30只,随机分为对照组(10只,给予常规饲料)和高脂组(20只,给予高脂饲料),共饲养10周。饲养期末取符合肥胖标准的16只大鼠定为肥胖组。检测2组大鼠空腹血糖(FBG)、胰岛素(Ins)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇指数(HDL-C)和低密度脂蛋白胆固醇(LDL-C)水平;采用稳态模型指数(HO-MA-IR)评价胰岛素抵抗程度;实时荧光定量RT-PCR检测2组大鼠肝脏FoxO1mRNA的表达。结果:饲养10周后,肥胖组大鼠FBG、Ins、TG、TC、LDL-C和HOMA-IR水平均较对照组升高(t分别为1.952、25.566、8.642、5.845、9.922和23.234,P均<0.05)。与对照组的(19.3±5.1)相比,肥胖组大鼠肝脏FoxO1mRNA表达量(35.6±4.4)增加(t=8.649,P<0.001)。肥胖组大鼠肝脏FoxO1mRNA表达与FBG(r=0.565,P=0.004)、Ins(r=0.564,P=0.004)、TG(r=0.626,P=0.001)和HOMA-IR(r=0.578,P=0.003)呈正相关。结论:高脂肥胖大鼠肝脏组织中FoxO1的表达量升高,且其与血糖、血脂代谢有关,可能参与肥胖和胰岛素抵抗的发病。 Aim:To explore the association of the expression of FoxO1 in liver with glycolipid metabolism in high-fat diet obese rats.Methods:Thirty male SD rats were randomly divided into 2 groups by taking routine(n=10) and high-fat diet(n=20).Levels of fasting blood glucose(FBG),serum insulin(Ins),triglyceride(TG),total cholesterol(TC),high density lipoprotein-C(HDL-C) and low density lipoprotein-C(LDL-C) were measured 10 weeks later.Insulin resistance was assessed by HOMA-IR.The FoxO1 mRNA expression level in liver was detected by real-time quantitative RT-PCR.Results:Levels of FBG,Ins,TG,TC,LDL-C and HOMA-IR were significantly higher in obese group than those in control group 10 weeks later(t=1.952,25.566,8.642,5.845,9.922,23.234,P0.05).The mRNA level of FoxO1 in liver of obese group(35.6±4.4) was higher compared with control group(19.3±5.1)(t=8.649,P0.001).The expression level of FoxO1 mRNA in liver was positively correlated with FBG(r=0.565,P=0.004),Ins(r=0.564,P=0.004),TG(r=0.626,P=0.001) and HOMA-IR(r=0.578,P=0.003).Conclusion:The increased expression of FoxO1 in liver of high-fat diet obese rats is associated with blood glucose and lipid levels.FoxO1 may be related to obesity and insulin resistance.
