摘要
目的:预测结核杆菌分泌抗原CFP21/MTB12/Rv3881c的HLA限制性细胞毒性T淋巴细胞(CTL)表位。方法:利用SYFPEITHI、BIMAS和NetCTL方法预测分析抗原CFP21/MTB12/Rv3881c的HLA-A*0201限制性CTL表位,进一步运用NetCTL方法对抗原CFP21/MTB12/Rv3881c的HLA-A其他等位基因和HLA-B限制性CTL表位进行预测分析。结果:初步筛选出3个候选抗原HLA-A*0201限制性CTL优势表位各4条,分别为CFP215-13、13-21、134-142、189-197;MTB1226-34、36-44、40-48、61-69;Rv3881c204-212、207-215、234-242、260-268。得到3个候选抗原HLA-A3和HLA-B7限制性CTL优势表位共21条。结论:初步筛选出抗原CFP21/MTB12/Rv3881c潜在的HLA-A*0201、HLA-A3、HLA-B7限制性表位33条。
Aim:To predict the HLA restricted cytotoxic T lymphocyte(CTL) epitopes in Mycobecterium tuberculosis CFP21/MTB12/Rv3881c.Methods:HLA-A0201 restricted CTL epitopes in CFP21/MTB12/Rv3881c were predicted by SYFPEITHI,BIMAS and NetCTL.The restricted T cell epitopes of HLA-B and the other alleles of HLA-A in CFP21/MTB12/Rv3881c were predicted by NetCTL.Results:Derived from the primary prediction results,the 12 predominant HLA-A0201 restricted T cell epitopes of the three candidates were CFP21 5-13,13-21,134-142,189-197,MTB12 26-34,36-44,40-48,61-69,and Rv3881c 204-212,207-215,234-242,260-268.And 21 predominant HLA-A3,B7 restricted T cell epitopes of the three candidates were also confirmed.Conclusion:Thirty three HLA-A0201,HLA-A3 and HLA-B7 restricted CTL epitopes derived from CFP21/MTB12/Rv3881c were predicted.
