摘要
目的探讨血小板膜糖蛋白(GP)Ⅰa基因和Ⅰbα基因多态性与脑梗死发生的关系,为缺血性脑卒中的预防及治疗提供理论基础。方法选择经CT或MRI证实的脑梗死患者302例(脑梗死组)和健康体检者196例(对照组);采用PCR-RFLP方法检测GPⅠa C807T基因与Ⅰbα基因HPA-2、Kozak序列多态性在2组中的分布频率。结果脑梗死组GPⅠa C807T等位基因频率明显高于对照组,差异有统计学意义(P<0.05);脑梗死组GPⅠbα基因Kozak序列C等位基因频率明显高于对照组差异有统计学意义(25.33% vs 10.20%,P<0.05);脑梗死组GPⅠbα基因HPA-2序列等位基因频率、基因型与对照组比较,差异无统计学意义(P>0.05)。结论 GⅠbα基因HPA-2序列多态性与脑梗死无相关性;GPⅠa C807T等位基因和Ⅰbα基因Kozak序列多态性可能是脑梗死的遗传危险因素。
Objective To explore the relationship between the Ⅰ a gene and Ⅰ ba gene polymorphism of platelet membrane glycoprotein(GP) and cerebral infarction in Chinese in the aspect of molecular etiology,in order to provide the theoretical basis for prevention and therapy of ischemic stroke. Methods GP Ⅰ a C807T genotype and GP Ⅰ ba genotype were investigated using PCR-RFLP in 302 patients with cerebral infarction and 196 control subjects. Results The frequencies of GP Ⅰ a C807T allele were significantly higher in the infarction group than in the control one (P (0.05). The Kozak genotype of GP Ⅰ ba with C allele was 25.33% in the cerebral in- farction group compared with 10.20 % in the control group, with statistically significant difference (P 〈0.05). There were no significant differences in genotype frequencies and allele frequencies of the HPA-2 genotype of GP Ⅰ ba between cerebral vascular thrombosis patients and controls (P 〉0.05). Conclusions The polymorphism of HPA-2 genotype of GP Ⅰ ba is not associated with cerebral vascular thrombosis in present cohort. Polymorphism of GP Ⅰ a C807T genotype and Kozak genotype of GP Ⅰ ba may be related to the genetic susceptibility of cerebral vascular thrombosis.
