摘要
目的:体外研究选择性环氧酶-2抑制剂塞来昔布(Celecoxib)与氟尿嘧啶(5-FU)联用对人胃癌细胞系SGC7901细胞增殖的相互作用。方法:采用MTT法观察不同浓度Celecoxib、5-FU单独和联合应用对SGC7901细胞生长的抑制作用,并使用中效原理判定两药合用的效果。结果:两种药物单独应用时,对细胞的抑制作用呈明显的剂量效应依赖关系;Celecoxib的中效浓度(IC50)为37.537μmol.L-1,5-FU的中效浓度为133.487μmol.L-1,这两种药物联用时存在"加速抑制作用"。应用中效原理分析显示,两种药物联用时存在协同效应,而与药物应用的先后顺序无关。结论:Celecoxib与5-FU在联合应用时的相互作用为协同效应,研究结果对提高胃癌的治疗效果具有一定的临床参考价值。
OBJECTIVE: To observe the effects of celecoxib(a selective Cycloxygenase-2 inhibitor) and 5-fluorouracil(5-FU) on human gastric carcinoma cell line SGC7901 in vitro.METHODS: The inhibitory effects of celecoxib and 5-FU either in combination or alone on SGC7901 cells were measured by MTT and the combination effect of the two drugs was evaluated according to the median-effect principle.RESULTS: Both celecoxib and 5-FU showed concentration-dependent inhibitory effect.The IC50 of celecoxib was 37.537 μmol·L^(-1) and that of 5-FU was 133.487 μmol·L^(-1);the combination of celecoxib and 5-FU showed an accelerated inhibitory effect.Analyzed in accordance with median-effect principle,the two agents were synergistic when used concurrently;however,the synergistic effect was independent of the administration order of the two agents.CONCLUSION: The interaction of celecoxib and 5-FU was proved to be synergistic.The results have clinical value for the improvement of the therapeutic outcome and in the treatment of gastric carcinoma.
