摘要
目的:研究受体胸腺内注射Fas抗体对心脏移植大鼠Fas/FasL表达的影响。方法:应用大鼠同种异位腹腔心脏移植模型,将模型动物随机分为三组,每组8只动物。A组:移植前后不给予处置;B组:给环孢素A,按每日8mg/kg于术前1d口服;C组:胸腺注射Fas抗体,以0.1mg/kg剂量在术前注射。以免疫组化法和逆转聚合酶链式反应(RT-PCR)法观察术后移植心脏局部的Fas及FasL的表达情况。结果:与A组相比,受体胸腺内注射Fas抗体可减少移植心脏局部的Fas/FasL基因表达(RT-PCR法半定量结果P<0.05)和蛋白表达(免疫组化法P<0.05)。结论:受体胸腺内注射Fas抗体是一种有效的抑制心脏移植急性排斥反应的方法,可使移植心肌局部Fas/FasL表达下调。
Objective:To investigate the influence of intrathymic injection of anti-Fas monoclonal antibody on Fas/FasL of rats after heart transplantation.Methods:Using rat allograft heterotopic heart transplantation models,rats were randomly divided into three groups:group A(control group),group B(was treated with cyclosporin before test),and group C(was treated with intrathymic injection of anti-Fas monoclonal antibody before test). The number of rats of every group was eight.After operations,the samples were observed the express of Fas/FasL on the transplanted heart by RT-PCR and immunohistochemical method.Results:Compared to group A,intrathymic injection of anti-Fas monoclonal antibody significantly decreased the gene express (RT-PCR,semiquantitative) and protein level (immunohistochemistry) of Fas/FasL on the transplanted heart (P〈0.05).Conclusion:Intrathymic injection of anti-Fas monoclonal antibody has a immunosuppressive effect on acute reject reaction and down regulation on the express level of Fas/FasL after rat heart transplantation.
