摘要
目的观察新加良附方对移植性人胃癌细胞(BGC-823)F/FL的表达影响。方法建立移植性人胃癌动物模型,并将动物模型随机分为胃癌模型对照组、西药5-FU对照组及新加良附方高、中、低剂量组,共5组。新加良附方大、中、小剂量组给药量分别为10g/kg、5g/kg和2.5g/kg;5-FU组给药剂量为17mg/kg;模型组给予等量无菌生理盐水。连续给药、给水15d处死模型小鼠分离肿瘤,瘤块称重,石蜡包埋,并将肿瘤组织切片,免疫组化法检测F/FL表达。结果新加良附方高剂量组能上调肿瘤组织F蛋白表达,下调FL蛋白表达,与模型对照组比较,有统计学意义(P〈0.05);新加良附方中剂量组能下调FL蛋白表达,与模型组比较,有统计学意义(P〈0.05)。结论新加良附方能通过启动F/FL介导的受体依赖型细胞凋亡途径,上调肿瘤组织F蛋白表达,下调FL蛋白表达而诱导胃癌细胞发生凋亡。
Objective To observe the effects of Xinjialiangfu formula (XJLFF) on F/FL expression of transplanted human gastric cancer cell ( BGC - 823 ). Methods Animal models with human gastric cancer xenografts were established and randomly divided into 5 groups ; namely model control group, 5 - FU group, XJLFF high - dose group, XJLFF medium - dose group and XJLFF low - dose group. XJLFF groups of high, medium and low dosage were administered XJLFF at the amount of 10 g/kg, 5 g/kg and 2.5 g/kg respectively; while 5 - FU group was administered 5 - FU at a dose of 17 mg/kg, and the model control group was given equivalent sterile normal saline. All mice were sacrificed after 15 days of continuous drug and water delivery. The tumor bilks were separated, weighed, embedded in paraffin, dissected into slices, and F/FL expression was detected by immunohistochemical method. Results F protein expression in tumor tissue of XJLFF high - dose group was up - regulated, while FL protein expression was down - regulated, and there were statistically significant difference compared with model control group ( P 〈 0. 05). In XJLFF medium - dose group, FL protein expression was down - regulated, and it was also significantly different from that in model control group ( P 〈 0. 05 ). Conclusion The X.ILFF can up - regulate F protein expression and down - regulate FL expression in tumor tissue by starting F/FL - mediated receptor - dependent apoptosis pathway, thereby inducing apoptosis in gastric cancer cells. This finding indicates that an in - depth research on the anti - tumor effect of XJLFF is in need and worthwhile.
