摘要
【目的】评价棉花纤维含糖检测中常用的糖分提取方法。水煮提取法、80%酒精提取法、0.05%TritonX-100溶液提取法。【方法】分别用这三种方法提取棉纤维中的糖分,并用蒽酮硫酸法测定可溶性总糖、用二硝基水杨酸法(DNS)测定还原糖来评价这三种提取方法。【结果】三种提取方法间所提取的含糖量结果差异不显著,但用水煮法制备的提取液测定出的可溶性糖和还原糖均稍高于其他两种提取方法。【结论】该方法耗时较短,成本低,因此建议选用水煮提取法,提取液可同时用于棉纤维的可溶性糖和还原性糖的分析。
[ Objective]There are three normal methods for extracting cotton fibre sugar, i. e. by boiling distill water, by 80% alcohol and by 0.05% Triton X- 100 solution. [Method]In this study, three methods were used to extract sugar from cotton fibre. Sulfuric acid - anthrone method was used to determine the content of the potysaccharide and 3,5 - dinitrosalicylic acid {DNS} method was used to analyze the content of reducing sugar in cotton fiber. [ Result]Variance analysis showed that there were no significant differences among the contents of the polysaccharide and reducing sugar by three extraction methods. However, the content of polysaceharide and reducing sugar extracted by boiling distill water was somewhat higher. [ Conclusion ] The extraction method was saving- time and lower cost, so this extraction method was suggested to be used during polysaeeharide and the reducing sugar analysis in cotton fiber.
