布鲁氏菌分子标记、毒力基因缺失株YZ-2生物学特性及免疫原性研究 被引量:3

Research on Biological Characteristics and Immunogenicity of the Molecular Marked and Virulence Deleted Vaccine Strain YZ-2 of Brucellosis

摘要 试验旨在考查布鲁氏菌分子标记、毒力基因缺失疫苗株YZ-2的生物学特性和免疫原性。对YZ-2菌落形态、生化特性、变异检查试验、毒力、遗传稳定性等生物学特性,以及免疫小鼠后体液免疫和细胞免疫水平进行试验,并与其亲本株S19进行比较。结果显示,YZ-2的形态及生化特性同亲本株S19相一致,但变异检查试验结果显示YZ-2由亲本株的光滑型变为粗糙型;毒力试验证实YZ-2的毒力与亲本株S19显著减弱,且YZ-2遗传稳定性良好。免疫原性试验证实,布鲁氏菌YZ-2在整体上具有与亲本株相似的免疫原性。结果表明,布鲁氏菌分子标记、毒力基因缺失疫苗株YZ-2的安全性较S19进一步提高,稳定性好,且具有与亲本株相近的免疫原性,有很好的应用前景。 To determine the biological characteristics and immunogenicity of the molecular and virulence deleted vaccine strain YZ-2 of Brucellosis. The biological characteristics of Brucella abortus YZ- 2 was evaluated by dyeing, toxicity, generation stability; vaccination of mice with YZ-2, the humoral immunity and cellular immunity were detected, and the results were analyzed. The research found that the biochemistry characteristics of YZ- 2 and S19 were nearly identical, and the colonies and the suspension have disparity. The variation experiment proved that YZ -2 had become rough type from smooth type. Virulent experiment suggested that strain YZ- 2 had lost some residual virulence of the parental strain, also YZ-2 has good heredity stability. Research verified strain YZ -2 has the immunogenicity as well as the parent strain. The results suggested that the safety of YZ-2 is better than parent strain, and the stability is well, has the same immunogenicity as parent strain; YZ-2 has good prospects to be used.
