摘要
目的:研究低密度脂蛋白(LDL)在荷瘤小鼠体内的分布及TNF、胆酸对其分布的影响。方法:将125I-LDL由荷瘤小鼠尾静脉注入,γ计数器测所取各组织放射活性并观察TNF和胆酸对其影响。结果:肿瘤组织对LDL的摄取量较高;TNF和胆酸均可降低肝、肾组织对LDL的摄取量(P<0.05);两者联用更可提高肿瘤组织对LDL的摄取量(P<0.05)。结论:肿瘤组织对LDL的摄取量较高;用药可以增加肿瘤组织对LDL的摄取量。提示LDL有可能作为一种良好的抗肿瘤药物载体。
Objective:This study examines the potential value of low density lipoprotein (LDL) as a vehicle for targeting cytotoxic drugs to tumor cells in S180-soft-bearing mouse model systems. Methods:The mice were injected with I 125 -LDL. 18 h later the animals were killed and sampled from a series of organs. We investigated the radioactivities assimilated by the tissues of liver,kidney,lung,spleen,heart,muscle,adrenal,gland,brain,bone,blood and tumor.Results:The uptake of LDL in tumor were higher than that of most studied tissuses except liver,kidney and adreal gland. TNF and bile-acid can diminish the concertration of LDL in liver and kidney, and combined administration of the two can enhance LDL focusing on tumor tissuses without affecting the others.Conclusion:LDL could be a potential vehicle of antitumor drug and the selectivity of LDL to tumor can be promoted by modification of reasonable medication.
