摘要
以湛江地区产量丰富的翡翠贻贝全脏器为原料,通过胰酶和枯草杆菌中性蛋白酶双酶水解、醇沉后得到翡翠贻贝糖胺聚糖粗品。使用二次回归正交旋转组合设计研究液料比、加酶量、酶解时间3个因素对糖胺聚糖粗品得率与糖胺聚糖含量乘积的影响,从而确定翡翠贻贝糖胺聚糖提取的最佳工艺条件。经响应面分析,回归模型具有较高的拟合度。结果显示,最佳提取工艺条件为液料比2.30:1,加酶量0.94%,酶解时间3.46h,在此条件下,重复实验结果与响应面法优化提取翡翠贻贝糖胺聚糖的模型预测值25.50(×10-4)之间的标准误差为1.10%,低于5%。
Glycosaminoglycan (GAG) was achieved from the whole viscera of Perna viridis via enzymatic hydrolysis by pancreatin and neutral protease of Bacillus subtilis and subsequent ethanol precipitation. The optimum extraction conditions for glycosaminoglycan from Perna viridis were determined by quadratic regression orthogonal rotational combining design and the optimal liquid-solid ratio, enzyme dosage and enzymolysis time were of 2.30:1, 0.94% and 3.46h, respectively. Under these conditions, the product of the yield of crude glycosaminoglycan and content ofglycosaminoglycan was close to the predicted value of 25.50 (×10^4) .
