摘要
[目的]探讨真姬菇单核体间RAPD标记多态性,为育种材料的选择提供一定的分子水平依据。[方法]以真姬菇(Hypsizygus marmoreus)工厂化栽培白色菌株SIEF3136的原生质体单核体和不同孢子单核体为材料,进行RAPD分析。[结果]筛选到38条多态性引物,共扩增341条条带,其中253条表现多态性,多态率为74.2%。31个菌株的相似系数为0.34~0.90。除单核体S18-16,同一交配型的单核体间的相似系数都在0.60以上,表现出较高的相似性。在大约0.61的相似水平上,所有菌株分为2组。PM-01与T1、T3聚为第1大类,PM-02与T2、T4聚为第2大类。另外,在不到0.62的水平上S18-16聚为第3大类,显示出极大的差异,这很可能是该单核发生突变所致。[结论]真姬菇子实体上担子在减数分裂中染色体交换很高,RAPD标记可以用于真姬菇单核多态性研究,并可为交配型鉴定提供参考。
[Objective]RAPD markers polymorphism of Hypsizygus marmoreus monokaryon strains was studied,so as to provide molecular basis for the selection of breeding material.[Method]Protoplast monokaryons and different spore monokaryons of the white cultivated strain SIEF3136 were selected as materials for RAPD analysis.[Result]38 primers were polymorphic between monokaryons.A total of 341 fragments were amplified,among them 253(accounting for 74.2%) were polymorphic.Genetic similarity coefficient matrix among 31 strains ranged from 0.34 to 0.90.Similarity coefficient of the same mating type monokaryons was 0.60 except monokaryon S18-16,which showed a higher similarity.At about 0.61 similarity level,all strains were divided into 2 groups.PM-01 with T1,T3 was clustered into the first category,and PM-02 with T2,T4 into the second category.In addition,S18-16 in less than 0.62 level was clustered into the third category,and it showed great difference,which was likely to be caused by mutation of the monokaryon.[Conclusion]Chromosome exchange in basidia of H.marmoreus fruitbody was high,and RAPD markers can be used polymorphism analysis among H.marmoreus monokaryons and can also provide reference for the mating type identification.
