摘要
目的研究丹参酮ⅡA(Tan ⅡA)联合顺铂(DDP)在体外对宫颈癌Hela细胞的凋亡作用及凋亡蛋白Bax与Bcl-2表达水平的变化。方法采用苏木素-伊红染色法观察细胞的形态学变化;流式细胞仪检测药物作用后的细胞凋亡情况;Western Blot测定Bax与Bcl-2蛋白在Hela细胞中的表达水平。结果对照组细胞未见凋亡,而两药单用组及联合组均可见凋亡细胞;除对照组外其他各组均可检测到细胞亚二倍体凋亡峰,Tan ⅡA+DDP组凋亡率高于两药单用组凋亡率,差别均有统计学意义(P<0.05);Bcl-2蛋白在各组中均有表达,但表达水平变化不明显(P>0.05);Bax蛋白在对照组中几乎不表达,而在各药物处理组中均有表达,且两药联合组中表达水平高于两药单用组,差别均有统计学意义(P<0.05)。结论Tan ⅡA与DDP二者联合可以增加Hela细胞凋亡率,其中通过增加Bax蛋白的表达,使Bax/Bcl-2比值升高,可能是其诱导细胞凋亡的机制之一。
Objective To investigate the effect of tanshinone IIA( Tan IIA) combined with cisplatin(DDP) on apoptosis of cervical cancer Hela cells and Bax/Bcl-2 protein expression of Hela cells in vitro. Methods The morphological changes were observed by hematoxylin and eosin staining;The apoptosis were detected by flow cytometry;Bax protein and Bcl-2 protein expression in Hela cells were detected by Western Blotting. Results There were no apoptotic cells in the control group,while the Tan IIA group and DDP group and the combined group were seen apoptotic cells. The sub-diploid apoptotic peak of cells were detected in each group except control group. The apoptotic rate in Tan IIA combined DDP group was significantly higher than that in Tan I1A group and DDP group( P 〈 0.05 ). The Bcl-2 protein expressed in each group, but there was no much difference among them( P 〉0.05). Bax protein expressed in each group excetp the control group and the expression levels in Tan IIA combined DDP group were significantly higher than thoes in Tan IIA group and DDP group ( P 〈 0.05 ). Conclusion Tan IIA combined DDP could induce cell apoptosis. The mechanisms of apoptosis-inducing may be associated with the increase protein Bax expression.
