青花菜高频再生体系建立及芽再生机理的研究 被引量:2

摘要 以青花菜无菌苗的不同部位为外植体,研究了青花菜离体再生体系的建立以及芽形态建成过程中其组织内部生理生化的变化。结果表明:在所选用的外植体中,萌发10d的无菌苗下胚轴和生根15d的试管苗茎段为再生的最适外植体;在研究所涉及的生长调节剂中以6-BA/IBA对芽分化最有效,当培养基中6-BA/IBA为4.0/0.3mg/L时,下胚轴和茎段外植体芽分化率均高达96.67%。愈伤组织形成、不定芽分化均与其组织内部大分子变化有关,在整个芽再生过程中可溶性蛋白质含量呈增加趋势,过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、多酚氧化酶(PPO)活性也随着芽的形态发生呈现相应的变化。 The regeneration of broccoli ( Brassica oleracea L. var. italica) in vitro, physiological and biochemical changes during morphogenesis of buds were studied. The results showed that hypocotyls and internodal segments taken separately from 10d -seedlings and 15d -regenerate test -tube plantlets were best explants for broccoli regeneration. 6 -BA/IBA was the most effective to bud- inducing in all the plant growth regulators used in this research. The highest bud differentiation rate 97% in the explants of both hypocotyls and internode was obtained when the concentration of 6 - BA/IBA was 4.0/0.3 mg/L. Calluses formation and buds differentiation were both related to changes of great molecules inside the tissues. The content of water soluble protein increased during the morphogenesis of buds. The activities of POD, CAT, SOD and PPO were also changed
